An Experimental Study of the Effect of Repeated Alcohol Administration on Halothane Metabolism and Hepatic Effect After Halothane Anethesia in Sprague-Dawley Rats


Abstract
The present study were performed to investigate the effects of halothane anesthesia on the metabolism and liver function in Sprague-Dawley rats pretreated with alcohol. Seventy two male Sprague-Dawley rats were divided into two groups. Saline
(5ml/kg,
in saline-treated group) or alcohol (400mg/kg, in alcohol-treated group) was injected through the tail vein for 7 days, and then exposed to 1% halothane(N2O-O2 3L/min-3L/min) 24 hours after the last treatment. Blood sampling was done before any
treatment through tail vein for control value. Intracardiac puncture for blood sampling was done under intraperitoneal injection of pentobarbital (50mg/kg) and hepatectomy was done to get the tissue sample for light microscopic examination 24
hour
after
exposure to halothane in one-half of the rats from each group: the remaining galf of the from each group were examined 96 hours after exposure to air or halothane. The liver function studies included alanine aminotransferase (ALT), aspartate
aminotrasferase (AST). Measurements of serum bromide and fluoride were done 24 or 96 hours after halothane anesthesia.
@ES The results were summarized as follows;
@EN 1. The liver function variables showed no significant statistical between salinetreated group and alcohol-treated group.
2. The histologic scores of light micorscopic findings showed no significant statistical differences between saline-treated group and alcohol-treated group.
3. Serum concentrations of bromide and fluoride after halothane anesthesia showed no significant statistical differences between saline-treated group and alcohol-treated group.
The above results concluded that alcohol administration for seven days does not affect the halothane metabolism and the hepatic effect of halothane anesthesia.
